Pigeonpea (Cajanus cajan L.), an important legume crop, plays a crucial role in global food security due to its high protein content, nitrogen-fixing ability, and adaptability to diverse climatic conditions. Despite its significance, accurate species identification and genetic characterization remain challenging due to morphological similarities among genotypes. DNA barcoding has emerged as a powerful tool for molecular characterization and species discrimination, providing a standardized method for genetic identification. This study aimed to develop and evaluate universal DNA barcoding primers for molecular characterization of pigeonpea genotypes using rbcL and matK genes and validating the application of DNA barcoding for accurate identification and genetic characterization of pigeon pea genotypes, offering a robust and efficient molecular approach for crop improvement and conservation programs.
